目的 建立鲫组织中复方磺胺甲噁唑含量的高效液相色谱检测方法, 并研究复方磺胺甲噁唑在鲫体内的残留消除规律。方法 按有效成分50 mg/kg体重的剂量对鲫进行复方磺胺甲噁唑口灌给药, 连续给药5天, 每天1次。并采集停药后1~15天期间的鲫性腺、肌肉、皮组织。通过二氯甲烷提取组织样品中的药物, 高效液相色谱-紫外检测法(HPLC-UV)测定药物含量。结果 TMP和SMZ最低检测限分别为0.015 μg/mL和0.006 μg/mL, 在0.05~100.00 μg/mL范围内线性关系良好(R2>0.9999), TMP和SMZ平均回收率分别为72%~81%和85%~92%。结论 该方法适合动物源性食品中复方磺胺甲噁唑残留量的检测。以水产养殖中复方磺胺甲噁唑正常使用剂量给药, TMP和SMZ在鲫可食性组织中分别于停药后14 d和2 d低于最高残留限量(50 μg/kg和100 μg/kg), 建议休药期为14天。 |

Objective To establish a method for the determination of the compound sulfamethoxazole by high performance liquid chromatography (HPLC) and research on the depletion of compound sulfamethoxazole residues in crucian carp (Carassius auratus). Methods After oral administration of compound sulfamethoxazole with a dosage of 50 mg/kg body weight per day for 5 d, once a day, the tissues of gonad, muscle and skin in crucian carp were collected from 1?15 d. Samples were extracted with dichloromethane and determined by HPLC. Results The lowest detection limit of TMP and SMZ were 0.015 μg/mL and 0.006 μg/mL. The method showed a good linearity for trimethoprim (TMP) and sulfamethoxazole (SMZ) in range of 0.05~100.00 μg/mL (R2>0.9999) and the recovery rates were 72%~81% and 85%~92%, respectively. Conclusion This method can be used for the determination of TMP and SMZ in aquatic products. At the 14 d and 2 d, the residues of TMP and SMZ were below the maximum residue limit (MRL) of China (50 μg/kg and 100 μg/kg) in the normal dosage. It suggested that the withdrawal time should be 14 d after administration by the normal dosage in crucian carp. |
