目的 建立一种快速、准确的RP-HPLC法测定胆固醇含量的方法。方法 样品在60 ℃水浴锅里磁力搅拌皂化45 min, 用石油醚提取后浓缩, 无水乙醇定容后, 用戴安C18色谱柱(4.6 mm×250 mm, 5 μm)进行分离, 甲醇作为流动相, 1 mL/min 流速进行等度洗脱。结果 方法的线性范围为10400 μg/mL, r=1.000, 检出限为20.6 μg/g, 相对标准偏差为0.66%2.12%, 回收率范围为92.50.2%。结论 该方法快速、准确、精密度高, 适用于食品中胆固醇的检测。 |

Objective To establish a rapid method for the determination of cholesterin in foods by RP-HPLC. Methods The samples were magnetically stirred and saponified in the water bath at 60 ℃ for 45 min, and extracted with petroleum ether and concentrated after making constant volume using anhydrous ethanol. Then, the samples were separated on a DIONEX C18 column (4.6 mm×250 mm, 5 μm) in elution with the mobile phase of methanol. Results The linearity of the method was 10400 μg/mL(r=1.000). The limit of detection was 20.6 μg/g with the relative standard deviations of 0.66%2.12%. The average recoveries were between 92.50.2%. Conclusion This method is rapid, accuracy, and precise, and it can meet the requirements of the cholesterol analysis. |
