      目的 建立灵芝三萜类化合物的快速、高通量定性的分析方法。方法 基于超高效液相色谱-四级杆-飞行时间质谱(Ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry, UPLC-Q-TOF-MS))技术高分离度、高分辨率的优势,通过对色谱分离和质谱检测条件进行优化,归纳三萜类化合物的质谱裂解规律及其特征碎片离子,构建灵芝三萜类化合物标准谱库;同时引入加标验证实验,结合加标样品、实际样品与标准品的保留时间及精确质荷比信息,实现实际样品中三萜类化合物快速鉴定。结果 结合标准品比对和三萜类裂解规律,共鉴定出32种三萜类组分和5种未知三萜组分。结论 该分析方法分离效果好、灵敏度高、专属性强,可实现灵芝三萜类化合物的快速定性与初步结构解析,适用于灵芝化学成分鉴定与质量控制,也为后续三萜类活性物质开发研究提供了方法参考。

      Objective To establish a rapid and high-throughput qualitative analysis method for Ganoderma triterpenoids. Methods Based on the advantages of ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) in high separation efficiency and high resolution, the chromatographic separation and mass spectrometric detection conditions were optimized. The mass spectral fragmentation patterns and characteristic fragment ions of triterpenoids were summarized, and a standard spectral library of Ganoderma triterpenoids was constructed. Meanwhile, sample spiking verification experiments were introduced. Combined with the retention time and accurate mass-to-charge ratio information of spiked samples, actual samples and reference standards, the rapid identification of triterpenoids was realized. Results Combined with standard substance comparison and triterpenoid cleavage rules, a total of 32 triterpenoid components and 5 unknown triterpenoid components were identified. Conclusion This analytical method has excellent separation performance, high sensitivity and strong specificity. It enables rapid qualitative identification and preliminary structural elucidation of Ganoderma triterpenoids. It is applicable to the chemical composition characterization and quality control of Ganoderma lucidum and also provides a methodological reference for further research of triterpenoid active substances.
