| 目的 推动具有缓解高尿酸血症(Hyperuricemia, HUA)作用的乳酸片球菌 TC-4 从研究走向应用、实现产业化发展,开展其培养基组分及冻干保护剂配方的优化研究。方法? 采用单因素试验、Plackett-Burman 试验与响应面法相结合,对乳酸片球菌 TC-4培养基的成分、浓度以及冻干保护剂的配方展开筛选。结果 最优培养基成分组合:葡萄糖26 g/L、酵母浸粉30 g/L,乙酸钠8 g/L、柠檬酸氢二铵2 g/L、碳酸氢钾2 g/L、硫酸镁0.4 g/L、硫酸锰0.075 g/L、吐温80 1 g/L,活菌数由3.5×109 CFU/mL提高至1.09×1010 CFU/mL,是优化前的3.11倍。最优冻干保护剂配方:L-谷氨酸钠8%,麦芽糖醇14%,脱脂乳粉18%,存活率达到90.77%。结论? 完成乳酸片球菌TC-4的培养基优化,成功制备高活菌制剂,为该菌株的产业化推进奠定坚实理论基础。 |
