      目的 推动具有缓解高尿酸血症(Hyperuricemia, HUA)作用的乳酸片球菌 TC-4 从研究走向应用、实现产业化发展,开展其培养基组分及冻干保护剂配方的优化研究。方法? 采用单因素试验、Plackett-Burman 试验与响应面法相结合,对乳酸片球菌 TC-4培养基的成分、浓度以及冻干保护剂的配方展开筛选。结果 最优培养基成分组合:葡萄糖26 g/L、酵母浸粉30 g/L,乙酸钠8 g/L、柠檬酸氢二铵2 g/L、碳酸氢钾2 g/L、硫酸镁0.4 g/L、硫酸锰0.075 g/L、吐温80 1 g/L,活菌数由3.5×109 CFU/mL提高至1.09×1010 CFU/mL,是优化前的3.11倍。最优冻干保护剂配方:L-谷氨酸钠8%,麦芽糖醇14%,脱脂乳粉18%,存活率达到90.77%。结论? 完成乳酸片球菌TC-4的培养基优化,成功制备高活菌制剂,为该菌株的产业化推进奠定坚实理论基础。

      ABSTRACT:Objective To promote the application of Pediococcus acidilactici TC-4, which has the effect of alleviating hyperuricemia (HUA), from research to industrialization, and to carry out the optimization research on the components of its culture medium and the formula of freeze-drying protectants.
