| 目的 开发一种电化学发光(electrochemiluminescence, ECL)试纸条快速检测黄曲霉毒素B1 (aflatoxin B1, AFB1)。方法 在聚氯乙烯衬底表面图案化碳油墨电极, 并与免疫层析试纸条组装, 制得ECL试纸条。采用循环伏安法考察了ECL试纸条的电化学性能。同时, 合成了Ru-PEI/Ti3C2Tx@Au复合材料, 并与AFB1单克隆抗体(AFB1 monoclonal antibodies, Anti-AFB1)偶联作为ECL信号探针。通过ECL试纸条检测线上AFB1牛血清白蛋白偶联物与ECL信号探针之间的竞争免疫反应, 实现了AFB1的比色和ECL双信号检测。结果 ECL试纸条具有良好的稳定性和特异性, 在最佳条件下, 比色信号与AFB1浓度对数在0.1~100.0 ng/mL范围内呈现良好的线性关系, 检出限为32 pg/mL。ECL信号与AFB1浓度对数在1.0 pg/mL~100.0 ng/mL范围内呈现良好的线性关系, 检出限为319 fg/mL。ECL试纸条适用于玉米样品中AFB1检测, 加标回收率为92.6%~110.3%, 相对标准偏差为0.9%~6.2%。结论 制备的ECL试纸条可用于玉米样品中AFB1高灵敏可视化快检, 为真菌毒素高灵敏快速检测提供了新方法。 |

| Objective To develop an electrochemiluminescence (ECL) test strip for the detection of aflatoxin B1 (AFB1). Methods The ECL test strip was fabricated by patterning carbon ink electrode onto the surface of a polyvinyl chloride substrate and then assembling them with a lateral flow immunoassay strip. The electrochemical performance of the ECL test strip was investigated using cyclic voltammetry. Meanwhile, Ru-PEI/Ti3C2Tx@Au composites were synthesized and conjugated with AFB1 monoclonal antibodies (Anti-AFB1) to serve as ECL signal probes. Through the competitive immunoreaction between the AFB1-bovine serum albumin conjugate immobilized on the test line of the ECL test strip and the ECL signal probes, colorimetric and ECL signals were obtained. Results The fabricated ECL test strips exhibited good stability and specificity. Under optimal conditions, the colorimetric signal showed a good linear relationship with the logarithm of AFB1 concentration in the range from 0.1 ng/mL to 100.0 ng/mL, with a limit of detection of 32 pg/mL. In addition, the ECL signal showed a good linear relationship with the logarithm of AFB1 concentration in the range from 1.0 pg/mL to 100.0 ng/mL, with a limit of detection of 319 fg/mL. Furthermore, satisfactory recovery rates ranging from 92.6% to 110.3% were achieved by the proposed ECL test strips, along with relative standard deviations of 0.9% to 6.2%. Conclusion The developed ECL test strip enables highly sensitive and rapid detection of AFB1 in corn samples, providing a new perspective for rapid mycotoxin detection. |
