| 目的 建立电感耦合等离子体质谱法(inductively coupled plasma mass spectrometry, ICP-MS)同时测定昭通乌天麻中25种矿质元素含量的分析方法, 评价和识别不同县区昭通乌天麻中元素含量差异。方法 对昭通市8个县区39份乌天麻样品进行微波消解, 采用ICP-MS测定其中25种元素含量; 通过线性关系、检出限、定量限、重复性及加标回收率验证方法的有效性, 同时建立昭通乌天麻矿质元素的指纹图谱并利用统计学软件, 结合模式识别分析方法中的主成分分析(principal component analysis, PCA)和正交偏最小二乘判别分析(orthogonal partial least squares discriminant analysis, OPLS-DA)进行产地分析识别。结果 方法线性关系良好(r>0.995), 重复性(相对标准偏差为0.8%~7.1%)和准确性(加标回收率93.2%~106.5%)高; 测定结果表明昭通乌天麻中矿质元素含量丰富且高K低Na的元素特征明显, 通过指纹图谱确定K、Fe、Mn、Zn、Sr、Ba、Al元素为昭通乌天麻的特征元素; 利用PCA和OPLS-DA能够实现不同县区昭通乌天麻的初步区分, 筛选出Fe、Al、Mn、Sr、Ba、K、Zn 7种元素对不同产区昭通乌天麻的分类起到了关键作用。结论 ICP-MS能够同时测定昭通乌天麻中多种金属元素, 通过分析昭通乌天麻中矿质元素的含量及其变化规律, 为昭通乌天麻的矿质营养评价及质量安全风险等提供参考, 为其资源多元化开发利用、产品推广溯源等提供数据支撑。 |

| Objective To establish an analytical method for the simultaneous determination of 25 kinds of mineral elements in Zhaotong Gastrodia elata Blume by inductively coupled plasma mass spectrometry, and evaluate and distinguish the differences in element content among Zhaotong Gastrodia elata Blume samples collected from different counties and districts. Methods The 39 samples of Zhaotong Gastrodia elata Blume collected from 8 districts in Zhaotong City were subjected to microwave digestion, and the concentrations of 25 kinds of elements were determined by ICP-MS. The validity of the method was verified through linear relationships, limits of detection, limits of quantitation, repeatability and spiked recovery rates. A fingerprint profile of mineral elements in Zhaotong Gastrodia elata Blume was established, and statistical software was used in conjunction with principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) to perform origin analysis and identification. Results The results showed that the method exhibited good linearity (r>0.995), high repeatability (relative standard deviation of 0.8%–7.1%), and high accuracy (spiked recovery of 93.2%–106.5%); the analytical results indicated that Zhaotong Gastrodia elata Blume was rich in mineral elements and exhibited distinct elemental characteristics of high potassium and low sodium. Through fingerprint analysis, elements such as K, Fe, Mn, Zn, Sr, Ba, and Al were identified as characteristic elements of Zhaotong Gastrodia elata Blume; PCA and OPLS-DA could be used to achieve preliminary differentiation among Zhaotong Gastrodia elata Blume from different counties, with 7 kinds of elements (Fe, Al, Mn, Sr, Ba, K and Zn) identified as playing a key role in classifying samples from different production areas. Conclusion The inductively coupled plasma mass spectrometry method can simultaneously determine multiple metallic elements in Zhaotong Gastrodia elata Blume. By analyzing the content and variation patterns of mineral elements in Zhaotong Gastrodia elata Blume, this study provides a reference for the mineral nutritional evaluation and quality safety risk assessment of Zhaotong Gastrodia elata Blume, and offers data support for its diversified resource development and utilization, product promotion and traceability. |
