      目的 建立固相萃取-高效液相色谱法同时测定洛神花酒中12种有机酸(酒石酸、羟基柠檬酸、乳酸、柠檬酸、琥珀酸、富马酸、没食子酸、原儿茶酸、绿原酸、咖啡酸、丁香酸和阿魏酸)含量的分析方法。方法 采用混合型强阴离子交换(polymeric anion exchange, PAX)固相萃取小柱净化富集。以0.1%磷酸水溶液和甲醇为流动相进行梯度洗脱, 经过CAPECELL PAK ADME C18液相色谱柱(4.6 mm×250 mm, 5 μm)分离, 采用二极管阵列多波长检测。结果 12种有机酸得到有效分离; 且在各质量浓度线性范围内线性关系良好, 相关系数(r)均在0.9998以上; 平均加标回收率为82.78%~101.52%, 相对标准偏差为0.90%~4.79%; 检出限为0.00047~0.49392 mg/L, 定量限为0.00157~1.64640 mg/L。结论 该方法操作简便、分离效果好、数据准确、灵敏度高, 适用于洛神花酒中有机酸的定量定性检测。

      Objective To establish an analytical method for the simultaneous determination of 12 kinds of organic acids (tartaric acid, hydroxycitric acid, lactic acid, citric acid, succinic acid, fumaric acid, gallic acid, protocatechuic acid, chlorogenic acid, caffeic acid, syringic acid and ferulic acid) in Hibiscus sabdariffa L. wine by solid-phase extraction and high performance liquid chromatography. Methods Mixed-mode strong anion exchange polymeric anion exchange (PAX) solid-phase extraction cartridges were used for purification and enrichment. The samples were eluted with 0.1% formic acid in water and methanol as mobile phases and separated on a CAPECELL PAK ADME C18 liquid chromatography column (4.6 mm×250 mm, 5 μm), and a diode array multi-wavelength detection were employed. Results The 12 kinds of organic acids were effectively separated; there was a good linear relationship within the linear range of various mass concentrations, with correlation coefficients (r) all above 0.9998; the average spiked recovery rates ranged from 82.78%–101.52%, with relative standard deviations of 0.90%–4.79%. The limits of detection were 0.00047–0.49392 mg/L and the limits of quantitation were 0.00157–1.64640 mg/L. Conclusion This method is simple to operate, good separation effect, accurate data and high sensitivity, making it suitable for the quantitative and qualitative detection of organic acids in Hibiscus sabdariffa L. wine.
