| 目的 探索乳酸片球菌AUI2505和罗伊氏乳杆菌AUI2506对高血糖斑马鱼的血糖调节作用。方法 通过模拟胃肠液耐受性实验、药敏性实验、溶血能力、疏水性和自聚力实验探究2株乳酸菌的体外益生特性及安全性; 通过浸泡在3%葡萄糖溶液中诱导斑马鱼血糖代谢紊乱, 建立高血糖斑马鱼模型, 然后对斑马鱼进行菌株干预作用实验。干预作用完成时测定各组斑马鱼存活率、血糖、胰岛素、炎症因子及糖耐量。结果 菌株AUI2505和菌株AUI2506在模拟胃液和肠液中的存活率分别大于80%和40%, 对大多数抗生素表现敏感, 不溶血, 显示出良好的疏水率和自聚力。斑马鱼高血糖模型显示, 经AUI2505组和AUI2506菌悬液干预后, 各组幼鱼的存活率均正常, 高血糖斑马鱼的血糖、炎症因子水平均显著降低(P<0.05), 葡萄糖耐量显著改善(P<0.05); 此外, AUI2505组显示胰岛素水平显著下降(P<0.05)。结论 乳酸片球菌AUI2505和罗伊氏乳杆菌AUI2506具有显著降糖功效, 为Ⅱ型糖尿病的益生菌辅助治疗提供了理论依据。 |

| Objective To investigate the blood glucose regulatory effects of Pediococcus acidilactici AUI2505 and Limosilactobacillus reuteri AUI2506 on hyperglycemic Danio rerio. Methods The in vitro probiotic properties and safety of 2 strains of lactic acid bacteria were evaluated through simulated gastrointestinal fluid tolerance tests, antibiotic susceptibility assays, hemolytic activity assessment and analyses of hydrophobicity and auto-aggregation ability. A hyperglycemic Danio rerio model was established by immersing the fish in water containing 3% glucose to induce blood glucose metabolism disorder, after which the Danio rerio were treated with the bacterial strains. After intervention, the survival rate, blood glucose levels, insulin levels, inflammatory factors and glucose tolerance of the Danio rerio in each group were measured. Results Strains AUI2505 and AUI2506 exhibited survival rates greater than 80% in simulated gastric fluid and greater than 40% in simulated intestinal fluid, respectively. Both strains were sensitive to most antibiotics tested and non-hemolytic. The 2 strains exhibited favorable hydrophobicity and auto-aggregation capacity. In the hyperglycemic Danio rerio model, it was demonstrated that following intervention with bacterial suspensions of AUI2505 and AUI2506, all experimental groups exhibited normal larval survival rates, blood glucose and inflammatory cytokine levels were significantly decreased (P<0.05), and glucose tolerance was significantly enhanced (P<0.05). Additionally, a significant reduction in insulin level was shown in the AUI2505 group (P<0.05). Conclusion Pediococcus acidilactici AUI2505 and Limosilactobacillus reuteri AUI2506 have significant hypoglycemic effects, providing a theoretical foundation for their use in adjunctive probiotic treatment for type 2 diabetes mellitus. |
