| 目的 优化复方黄精膏功效成分的提取工艺, 并评价其免疫调节活性。方法 以粗多糖、总皂苷、虫草素含量和干膏率为评价指标, 通过层次分析法(analytic hierarchy process, AHP)-客观赋权法(criteria importance through intercriteria correlation, CRITIC)混合加权法和Box-Behnken设计-响应面法对提取工艺参数进行优化; 采用碳廓清实验、迟发型变态反应实验和血清血溶素实验以评价其免疫调节活性。结果 复方黄精膏功效成分的最佳提取工艺为提取时间117 min, 提取次数3次, 提取温度94 ℃, 料液比1:14 (g:mL)。复方黄精膏能增强小鼠迟发型变态反应能力, 提高单核-巨噬细胞的吞噬能力和血清溶血素的含量。结论 该工艺合理可行, 最佳工艺下提取的复方黄精膏具有良好的免疫调节活性, 为复方黄精膏的进一步开发与应用提供了参考。 |

| Objective To optimize the extraction process of the functional components of compound Polygonati rhizoma ointment and evaluate its immune regulating activity. Methods Using crude polysaccharides, total saponins, cordycepin content and dry extract rate as evaluation indicators, the extraction process parameters were optimized through analytic hierarchy process (AHP)-criteria importance through intercriteria correlation (CRITIC) mixed weighting method and Box-Behnken design response surface methodology. Carbon clearance test, delayed type hypersensitivity test and serum hemolysin test were used to evaluate its immune regulating activity. Results The optimal extraction process for the functional components of compound Polygonati rhizoma ointment was 117 min of extraction time, 3 times of extraction, extraction temperature of 94 ℃ and a solid-liquid ratio of 1:14 (g:mL). Compound Polygonati rhizoma ointment could enhance the delayed type hypersensitivity ability of mice, improve the phagocytic ability of monocytes macrophages and increase the content of serum hemolysin. Conclusion The process is reasonable and feasible, and the compound Polygonati rhizoma ointment extracted under the optimal process has good immune regulatory activity, providing a reference for the further development and application of compound Polygonati rhizoma ointment. |
