      目的 建立低共熔溶剂(deep eutectic solvents, DESs)-甲醇-水提取的超高效液相色谱-串联质谱法测定云南省特色的云南铁皮石斛和紫皮石斛中7种活性成分的方法, 对云南的特色云南铁皮石斛和紫皮石斛进行鉴别研究。方法 样品经DESs-甲醇-水溶液冰浴下超声提取后, 采用超高效液相色谱-串联质谱法, 多反应监测模式测定, 化学计量学进行数据分析。结果 7种活性成分在一定质量浓度范围内的相关系数较好, 检出限和定量限的范围分别为0.006~0.030 mg/kg和0.020~0.100 mg/kg, 精密度、重复性和稳定性的相对标准偏差范围为0.95%~5.45%, 相对于铁皮石斛, 紫皮石斛在金圣草素、异槲皮素、芹菜素和原儿茶酸的含量上调, 圣草酚的含量下调, 而柚皮素和香草酸的含量在两种石斛中的差异不大。结论 方法的灵敏度和稳定性较好, 铁皮石斛和紫皮石斛可通过正交偏最小二乘判别分析进行有效的鉴别, 该研究结果可对云南省特色石斛的质量评价提供参考。

      Objective To establish a method for the determination of 7 kinds of active components in the characteristic Dendrobium officinale and Dendrobium devonianum in Yunnan Province by deep eutectic solvents (DESs)-methanol-water extraction and ultra performance liquid chromatography-tandem mass spectrometry, and conduct identification research on the characteristic Dendrobium officinale and Dendrobium devonianum in Yunnan Province. Methods After ultrasonic extraction of the samples in a DESs-methanol-water solution ice bath, ultra performance liquid chromatography-tandem mass spectrometry was employed for analysis in multiple reaction monitoring mode, with chemometric data analysis conducted. Results The correlation coefficients for the 7 kinds of active ingredients within a certain mass concentration range were relatively good. The limits of detection and limits of quantitation ranged from 0.006 mg/kg to 0.030 mg/kg and 0.020 mg/kg to 0.100 mg/kg, respectively. The relative standard deviations ranged for precision, repeatability and stability were 0.95% to 5.45%. Compared to Dendrobium officinale, Dendrobium devonianum exhibited higher levels of ginsenoside, isoquercitrin, apigenin and protocatechuic acid, while showing lower levels of ginsenoside; the content of naringin and vanillic acid remained relatively consistent between the 2 kinds of species. Conclusion The sensitivity and stability of the method are relatively good. Dendrobium officinale and Dendrobium devonianum can be effectively discriminated by orthogonal partial least squares discriminant analysis. The research results can provide a reference for the quality evaluation of the characteristic dendrobium in Yunnan Province.
