目的 采用超高效液相色谱-串联质谱法(Ultra-High Performance Liquid Chromatography-Tandem Mass Spectrometry,UHPLC-MS/MS),对新疆不同地区的孜然进行硒代氨基酸含量分析。方法 样品采用水提取法,0.1%的乙腈(A)和水(B)二元流动相体系、多反应监测模式进行监测,基质匹配外标法进行定量,对硒代胱氨酸、硒代蛋氨酸和Se-(甲基)硒基-L-半胱氨酸等3种硒代氨基酸进行色谱分离、测定。结果 北疆种植孜然的硒代氨基酸含量分布最高,为1.2~6.0 μg/kg,其次是全疆0.8~4.0 μg/kg;东疆和南疆的含量相似,在0.8~4.0 μg/kg,其中产自吐鲁番的孜然中3种硒代氨基酸含量较高。环节中硒代氨基酸总含量的大小顺序为批发环节>零售超市环节>农贸环节,3种硒代氨基酸中硒代胱氨酸含量最多,占比最大。结论 本研究采用水提法以及UPLC-MS/MS分析3种硒代氨基酸测定结果准确且操作便捷。结果分析各含量显示同一区域内所产孜然中硒代氨基酸的含量和质量仍可能呈现较大差异,在不同环节下的各种物质含量有明显差异,其中批发环节保持了硒代氨基酸的稳定性,3种硒代氨基酸含量较高。以期对孜然中硒代氨基酸高效测定提供数据支持。 |

Objective The content of selenoamino acids in cumin from different regions of Xinjiang was analyzed using ultra-high performance liquid chromatography-tandem mass spectrometry. Method Using the water extraction method, a binary mobile phase system consisting of 0.1% acetonitrile (A) and water (B), and multiple reaction monitoring mode for detection, matrix-matched external standard method for quantification, chromatographic separation and determination were performed for three seleno-amino acids: selenocysteine, selenomethionine, and Se-(methyl)selenocysteine.Result The distribution of seleno-amino acid content in cumin grown in the northern region is the highest, ranging from 1.2 to 6.0 μg/kg, followed by the entire region with 0.8 to 4.0 μg/kg; the content in the eastern and southern regions is similar, ranging from 0.8 to 4.0 μg/kg, with cumin from Turpan having a higher content of the three seleno-amino acids. The total content of seleno-amino acids in the different links is in the order of wholesale links > retail supermarket links > agricultural market links, with selenocysteine being the most abundant among the three seleno-amino acids, having the largest proportion.Conclusion This study employed water extraction and Ultra-High Performance Liquid Chromatography-Tandem Mass Spectrometry (UHPLC-MS/MS) analysis for the accurate and convenient determination of three seleno-amino acids. The results showed that within the same region, the content and quality of seleno-amino acids in cumin can still vary significantly. There are also clear differences in the content of various substances at different stages, with the wholesale stage maintaining the stability of seleno-amino acids, and the content of the three seleno-amino acids being relatively high. The aim is to provide data support for the efficient determination of seleno-amino acids in cumin. |
