目的 优化浊点萃取协同超声波辅助提取桑叶黄酮工艺, 并对其降血脂作用进行研究。方法 以桑叶黄酮得率为指标, 采用单因素和响应面实验, 优化浊点萃取协同超声波辅助法的提取工艺条件。同时将小鼠随机分为桑叶黄酮低、中、高剂量组、正常对照组、模型对照组和辛伐他汀组。除正常对照组外, 其余5组喂养60%高脂饲料构建高血脂小鼠模型。测定小鼠的体重、血清中血清总胆固醇(total cholesterol, TC)、甘油三酯(triglyceride, TG)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol, LDL-C)和高密度脂蛋白胆固醇(high density lipoprotein cholesterol, HDL-C)含量及肝组织中超氧化物歧化酶(superoxide dismutase, SOD)活性、丙二醛(malondialdehyde, MDA)含量和谷胱甘肽过氧化物酶(glutathione peroxidase, GSH-Px)活性。结果 最佳提取工艺条件为: 十二烷基硫酸钠添加量2.1%、超声波功率400 W、超声时间42 min, 此时桑叶黄酮得率2.82%, 此法的提取效果优于其他两种单一的提取方式。桑叶黄酮对高血脂小鼠降血脂实验结果得出: 桑叶黄酮能有效抑制高血脂小鼠体重的增加, 减少高血脂小鼠血清中TC、TG、LDL-C的含量, 降低高血脂小鼠肝组织中MDA含量, 提高GSH-Px和SOD活性, 表明桑叶黄酮对高血脂小鼠具有良好的降血脂作用。结论 优化后的提取条件能够有效提取桑叶黄酮, 得到的桑叶黄酮具有较好的降血脂作用, 该研究可为桑叶黄酮的提取及开发应用提供参考。 |

Objective To optimize the extraction process of mulberry leaf flavonoids by cloud point extraction assisted ultrasonic extraction and study its hypolipidemic effect. Methods Using the yield of mulberry leaf flavonoids as an indicator, single factor and response surface methodology were used to optimize the extraction process conditions of cloud point extraction assisted ultrasonic extraction method. At the same time, mice were randomly divided into low, medium and high dose groups of mulberry leaf flavonoids, normal control group, model control group and simvastatin group. Except for the normal control group, the other 5 groups were fed 60% high fat feed to construct a hyperlipidemic mouse model. The body weight, serum total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C) content of serum, superoxide dismutase (SOD) activity, malondialdehyde (MDA) content and glutathione peroxidase (GSH-Px) activity of liver tissue were measured. Results The optimum extraction conditions were: Sodium dodecyl sulfate addition was 2.1%, ultrasonic power was 400 W, and ultrasonic time was 42 minutes. Under these conditions, the yield of mulberry leaf flavonoids was 2.82%, and the extraction effect of this method was superior to the other 2 single extraction methods. The results of the experiment on lowering blood lipids in hyperlipidemic mice with mulberry leaf flavonoids showed that mulberry leaf flavonoids could effectively inhibit the increase of body weight, reduce the content of TC, TG, LDL-C in the serum of hyperlipidemic mice, reduce the content of MDA in the liver tissue of hyperlipidemic mice, increase the activity of GSH-Px and SOD, indicating that mulberry leaf flavonoids had good hypolipidemic effect on hyperlipidemic mice. Conclusion The optimized extraction conditions can effectively extract mulberry leaf flavonoids, and the obtained mulberry leaf flavonoids have good hypolipidemic effect, this study can provide reference for the extraction, development, and application of mulberry leaf flavonoids. |
