目的 通过非靶向代谢组学分析宁杞7号、宁杞9号、宁杞10号3个不同品种的宁夏枸杞的差异性代谢物。方法 采用超高效液相色谱-四极杆/静电场轨道阱高分辨质谱法建立非靶向代谢组学研究方法, 通过主成分分析(principal component analysis, PCA)、偏最小二乘法分析(partial least-squares discrimination analysis, PLS-DA)模型、聚类分析等统计学方法对不同品种的宁夏枸杞进行差异分析。结果 宁杞7号、宁杞9号、宁杞10号3种宁夏枸杞品种中正、负模式下均存在的代谢物有393个, 共鉴定出17种差异性代谢物[变量投影重要性(variable importance in project, VIP)>1], 包括有机酸类及其衍生物3种、氨基酸类及其衍生物2种、黄酮类及黄酮醇类化合物7种、酚酸类2种、其他3种。结论 该研究确定了宁杞7号、宁杞9号、宁杞10号3种宁夏枸杞品种的差异代谢物, 可以为不同宁夏枸杞品种的差异性分析提供新的研究思路, 对优质宁夏枸杞种质资源的挖掘与选育提供重要参考。 |

Objective To explore the differences of metabolites among 3 varieties of Lycium barbarum L., including Ningqi No.7, Ningqi No.9 and Ningqi No.10 by non-targeted metabolomics. Methods The non-targeted metabolomics research method was established by ultra performance liquid chromatography-quadrupole-electrostatic field orbitrap high-resolution mass spectrometry. The difference analysis of different varieties of Lycium barbarum L. was conducted by principal component analysis (PCA), partial least squares discrimination analysis (PLS-DA) model, cluster analysis and other statistical methods. Results The 393 metabolites in both positive and negative modes were identified in Ningqi No.7, Ningqi No.9 and Ningqi No.10, and 17 kinds of different metabolites including 3 kinds of organic acids and their derivatives, 2 kinds of amino acids and their derivatives, 7 kinds of flavonoids and flavonols, 2 kinds of phenolic acids and 3 kinds of other compounds were screened variable importance in project (VIP)>1 from Ningqi No.7, Ningqi No.9, Ningqi No.10. Conclusion This study determines the differential metabolites of 3 varieties of Lycium barbarum L., including Ningqi No.7, Ningqi No.9 and Ningqi No.10, and can provide a new research direction for the difference analysis of different varieties Lycium barbarum L, and provide an important reference for the mining and breeding the high quality germplasm resources of Lycium barbarum L. in the later period. |
