      目的 采用超高效液相色谱-四极杆-静电场轨道离子阱高分辨质谱法(ultra performance liquid chromatography-quadrupole-electrostatic field orbitrap high resolution mass spectrometry, UPLC-Q-Orbitrap HRMS)建立防城金花茶和越南金花茶叶主要化学成分的分析与鉴定方法。方法 样品采用ACQUITY UPLC? HSS T3色谱柱分离, 以0.1%甲酸乙腈-0.1%甲酸水溶液为流动相, 流速0.300 mL/min, 柱温35℃, 梯度洗脱, 在正负离子模式下采集数据, 根据高分辨质谱提供的精确相对分子质量和碎片离子信息, 结合ChemSpider、mzVault、mzCloud质谱数据库和相关文献报道对金花茶叶的化学成分进行分析。结果 从金花茶叶中共确认180个化学成分, 包含了28个黄酮类化合物, 9个苯丙素类化合物, 15个萜类化合物, 3个生物碱类化合物和128个其他类成分。结论 应用UPLC-Q-Orbitrap HRMS结合数据库可实现对金花茶主要化学成分的分析鉴定, 为进一步研究金花茶的全面质量控制提供了科学依据。

      Objective To analyze and identify the main chemical components in the leaves of Camellia nitidissima Chi and Camellia nitidissima C.W.Chi by ultra performance liquid chromatography-quadrupole-electrostatic field orbitrap high resolution mass spectrometry (UPLC-Q-Orbitrap HRMS). Methods The samples were loaded on the ACQUITY UPLC? HSS T3 column for gradient elution with 0.1% formic acid in acetonitrile-0.1% formic acid aqueous solution as mobile phase at a flow rate of 0.300 mL/min. The column temperature was 35℃. The datas were acquired in positive and negative ion modes. The chemical constituents were analyzed according to the precise relative molecular mass and fragment ion information provided by high resolution mass spectrometry, combined with ChemSpider, mzVault, mzCloud mass spectrometry databases and related literature reports. Results A total of 180 chemical constituents were isolated and identified, consisting of 28 flavonoids, 9 phenylpropanoids, 15 terpenoids, 3 alkaloids and 128 other components. Conclusion The combination of UPLC-Q-Orbitrap HRMS and databases can realize the analysis and identification of main chemical components in Camellia nitidissima, so as to provide a scientific basis for further research on the overall quality control of Camellia nitidissima.
