目的 优化碱水提取法提取江蓠(Gracilaria asiatica)中活性多糖的工艺。方法 以江蓠多糖的得率为指标, 采用碱水提取法从江蓠中提取多糖, 以碱浓度、提取温度和料液比作为单因素变量, 在单因素试验的基础上, 采用响应面法优化了提取江蓠多糖的工艺条件。结果 碱水提取法提取江蓠多糖的最佳工艺条件为: 碱浓度0.03 mol/L、提取温度93.00 ℃和料液比1:126.00 (g/mL), 在此条件下, 江蓠多糖得率最高, 达到7.85%±0.56%, 与响应面预测值7.94%相近, 表明响应面模型是成功的、可行的。结论 本研究建立了碱水提取法提取江蓠多糖的工艺, 该工艺条件稳定可行, 得率较高, 可为江蓠多糖的分离和综合利用提供参考。 |

Objective To optimize the extraction technology of active polysaccharides from Gracilaria asiatica by alkali water extraction method. Methods Taking the yield of polysaccharide as evaluation indicators, the polysaccharides from Gracilaria asiatica were obtained using alkali water extraction method. The alkali solution concentration, extraction temperature, and solid-liquid ratio were selected as single factor variables. Based on the single factor experiments, the extraction technology conditions of Gracilaria asiatica polysaccharides were optimized by response surface method. Results The optimum technological conditions for extracting Gracilaria asiatica polysaccharides by alkaline water extraction were as follows: Alkali solution concentration 0.03 mol/L, extraction temperature 93.00 ℃, and solid-liquid ratio 1:126.00 (g/mL). Under the conditions, the yield of Gracilaria asiatica polysaccharide was the highest, reaching 7.85%±0.56%, which was close to the predicted value of 7.94%, indicating that the applied response surface model was successful and feasible. Conclusion The technology of alkali water extraction of polysaccharide from Gracilaria asiatica is established, the technology is stable and feasible, and has higher yielding rate, which can provide a reference for the separation of polysaccharides from Gracilaria asiatica and the comprehensive utilization of them. |
