      目的 建立高效液相色谱法检测分析马齿苋中腺苷含量的分析方法。方法 比较纯水、不同体积分数的乙醇、不同体积分数的甲醇为提取溶剂时, 马齿苋中腺苷提取量差异, 优选最佳的提取方式。采用安捷伦ZORBAX SB-Aq色谱柱(4.6 mm×250 mm, 5 μm), 以乙腈:0.1%乙酸=7:93(V/V)为流动相, 流速0.5 mL/min, 检测波长260 nm, 柱温40℃, 对马齿苋中的腺苷进行测定。结果 纯水提取得到的腺苷含量高于不同体积分数的乙醇和甲醇提取。腺苷在1.71~171.0 mg/L范围内线性关系良好, 相关系数r2=0.9999, 加样平均回收率为92.65%。结论 该方法结果可靠, 操作简单, 可以准确的测定马齿苋中的腺苷含量, 可以作为马齿苋质量控制的方法之一。

      Objective To establish a method for the determination of adenosine in Portulaca oleraceaby high performance liquid chromatography. Methods Using pure water, different volume fractions of ethanol and different volume fractions of methanol as extraction solvents, thedifference of extraction amount of adenosine in Portulaca oleracea L. was compared, and the best extraction method was optimized. Adenosine in Portulaca oleracea L. was determined on Agilent ZORBAX SB-Aq column (4.6 mm×250 mm, 5 μm) with acetonitrile: 0.1% acetic acid=7:93 (V/V) as mobile phase, flow rate 0.5 mL/min, detection wavelength 260 nm and column temperature 40 ℃. Results The content of adenosine extracted by water was higher than that extracted by ethanol and methanol with different volume fractions. The linear range of adenosine was 1.71-171.0 mg/L, the correlation coefficient r2=0.9999, and the average recovery rate was 92.65%. Conclusion This method is reliable, simple, can be used for the determination of adenosine in Portulaca oleracea L.and can be used as one of the methods for quality control of purslane.
