      目的 研究卵白蛋白(ovalbumin, OVA)对变应性鼻炎(过敏性鼻炎)小鼠模型的影响。方法 小鼠随机分为7组: 对照组、高浓度OVA模型+PBS组、中浓度OVA模型+PBS组、低浓度OVA模型+PBS组、高浓度OVA+阳性药组、中浓度OVA+阳性药组、低浓度OVA+阳性药组。采用OVA全身致敏及局部攻击相结合的方法构建变应性鼻炎小鼠模型, 以PBS、丙酸氟替卡松(0.008 mg/kg)滴鼻, 观察小鼠鼻部抓挠、喷嚏数症状和脏器指数; ELISA法定量检测血清免疫球蛋白E(IgE)、白细胞介素-4(IL-4)、白细胞介素-10(IL-10)和干扰素-γ(IFN-γ)的含量; 制备脾细胞悬液对比脾细胞悬液CD4+CD25+T细胞变化。结果 建模成功后, 高浓度OVA+阳性药组与高浓度OVA模型+PBS组相比, 血清IgE、IL-10、IL-4、IFN-γ水平和CD4+CD25+T细胞水平变化不明显, 差异无统计学意义(P>0.05); 中、低浓度OVA+阳性药组与中、低浓度OVA模型+PBS组相比, 血清IgE、IL-10和IL-4水平显著降低, CD4+CD25+T细胞水平和IFN-γ水平显著升高, IFN-γ/IL-4比值升高, 差异有统计学意义(P<0.001或P<0.05)。结论 卵白蛋白的浓度选择对变应性鼻炎模型影响较大, 致敏用卵白蛋白为中浓度时对阳性药的检测效率较高。

      Objective To study the effect of ovalbumin (OVA) on allergic rhinitis in mice. Methods The mice were randomly divided into 7 groups: control group, high concentration OVA model+PBS group, medium concentration OVA model+PBS group, low concentration OVA model+PBS group, high concentration OVA+positive drug group, medium concentration OVA+positive drug group and low concentration OVA+positive drug group. The mice model of allergic rhinitis was established by the combination of systemic sensitization and local attack with OVA. The mice nose was dripped with PBS and fluticasone propionate (0.008 mg/kg). The symptoms of scratching and sneezing and organ index were observed. The contents of serum immunoglobulin E (IgE), interleukin-4 (IL-4), interleukin-10 (IL-10) and interferon-γ (IFN-γ) were measured quantitatively by ELISA. The changes of CD4+CD25+T cell in spleen cell suspension were observed. Results Compared with the high concentration of OVA+ PBS group, the levels of serum IgE, IL-10, IL-4, IFN-γ and CD4+CD25+T cells were significantly different from mesin the high concentration OVA+positive drug group had no significant difference (P>0.05); compared with the medium and low concentration OVA+PBS group, the levels of serum IgE, IL-10 and IL-4 in the medium and low concentration OVA+positive drug group were significantly lower, the level of CD4+CD25+T cell and IFN-γ increased significantly, the ratio of IFN-γ to IL-4 increased significantly (P<0.001 or P<0.05). Conclusion The concentration of ovalbumin has a great influence on the allergic rhinitis model. When the concentration of ovalbumin is medium, the detection efficiency of positive drugs is high.
