      目的 建立高效液相色谱法(high performance liquid chromatography, HPLC)测定奶茶饮料中茶黄素的含量。方法 奶茶样品经0.1%乙酸乙醇沉淀蛋白后, 5000 r/min离心5 min, 取上清液进行检测。采用 0.5%乙酸(A)和乙腈(B)作为流动相进行梯度洗脱, 二极管阵列检测器在280 nm处进行监测, 流速1.5 mL/min, 色谱柱温度40 ℃, 外标法定量。结果 本方法可以实现4种目标化合物的分离检测, 4种茶黄素的回收率在89.06%~96.51%范围内, 相对标准偏差小于5%(n=5)。结论 该方法前处理时间短、操作简便、稳定性好、准确度高, 可用于奶茶饮料中茶黄素的测定。
英文摘要:
      Objective To establish a method for determination of the content of theaflavins in milk tea beverages by high performance liquid chromatography (HPLC). Methods Milk tea samples were extracted with 0.1% acetic acid in ethanol solution, then centrifuged at 5000 r/min for 5 min, and the supernatant was detected. The theaflavins in the extracts was separated by using a gradient elution program of 0.1% acetic acid aqueous solution (A) and acetonitrile solution (B), and monitored at 280 nm by diode array detector. The flow rate was 1.5 mL/min. The temperature of column was 40 ℃. The content was quantified with exterior standard. Results This method could realize the separation and detection of 4 target compounds. The recoveries were ranged from 89.06% to 96.51 % and the relative standard deviations (RSDs) were less than 5% (n=5). Conclusion The proposed method is fast, simple, stable and accurate, which is suitable for detecting theaflavins in milk tea beverage.
