目的 优化锥栗壳色素提取工艺, 为加工废弃物锥栗壳资源得到有效的开发利用提供参考。方法 以新鲜干燥的锥栗壳粉为原料, 采用恒温-回流法提取锥栗壳色素, 在单因素实验基础上, 固定液固比30:1(mL:g)、提取1次, 优化乙醇浓度、提取温度、提取时间3个因素, 获得更高色素得率, 利用Box-Benhnken中心组合设计和响应曲面分析法, 研究各个因素及其交互作用对色素得率的影响, 模拟获得二次多项式回归方程的预测模型。结果 锥栗壳色素的最佳提取工艺条件为: 乙醇浓度47%、提取温度90 ℃、提取时间2.5 h。在此条件下, 锥栗壳色素的得率为17.29%, 与预测值17.40%相对误差为0.63%。结论 通过响应面法优化得出的数学模型合理可靠, 获得的锥栗壳色素提取工艺可行。 |

Objective To optimize the extraction process of Castanea henryi shell pigment and provide reference for the effective development and utilization of the processing waste Castanea henryi shell resources. Methods Using the fresh and dry Castanea henryi shell powder as raw material, the Castanea henryi shell pigment was extracted by constant temperature-reflux method. Based on single factor experiment, liquid-solid ratio fixed 30:1(mL:g), extracted one time, three extraction parameters including ethanol concentration, extraction temperature and extraction time were optimized for achieving higher yield of the pigment. The interaction of the various factors and their influence on the extraction rate were studied by central composite design and response surface methodology and the prediction model of quadratic polynomial regression equation was obtained. Results The optimum extraction process of the Castanea henryi shell pigment was as follow: ethanol concentration was 47%, extraction temperature was 90 ℃ and extraction time was 2.5 h. Under this condition, the yield of the Castanea henryi shell pigment was 17.29%, compared to the predictive value 17.40%, the relative error of 0.63%. Conclusion The mathematical model optimized by the response surface method is reasonable and reliable, and the extraction process of Castanea henryi shell pigment is feasible. |
