目的 以黑果枸杞为原料, 研究黑果枸杞花青素提取的最佳工艺条件及其纯化工艺, 并对纯化效果进行检验。方法 采用单因素和正交实验, 以乙醇为提取溶剂, 对花青素进行提取, 对提取温度、提取时间、料液比和乙醇浓度等参数进行优化; 将提取液离心并蒸发浓缩后用大孔树脂进行纯化, 对纯化工艺进行研究, 确定最佳纯化工艺。结果 确定黑果枸杞花青素的最佳提取工艺为: 以pH3.0的75%乙醇溶液为提取溶剂, 提取温度为40 ℃, 提取时间为2 h, 料液比为1:40。在pH3.0, 温度低于50 ℃, 流速为1 mL/min的条件下, 大孔树脂对黑果枸杞花青素的最大吸附量达61.21 mg/mL, 花青素回收率为44.35%, 花青素含量为118 mg/g, 是纯化前的10.91倍, 通过冷冻干燥洗脱液得到外观呈黑紫色的花青素产品。结论 确定了黑果枸杞花青素乙醇浸提法的最佳工艺条件, 证明了AB-8大孔树脂具有良好的吸附能力和选择吸附性。 |

Objective To study on the optimum extraction conditions and purification process of anthocyanin from Lycium ruthenicum Murr (LR) by using LR as raw material, and verify the purification effect. Methods Basing on single factor and orthogonal experiments, the anthocyanin was extracted with ethanol, and the extraction parameters were optimized which included extracting temperature, extracting time, ratio of material to liquid and ethanol concentration. Then the extract was purified by macroporous resin after centrifugation and evaporation concentration, and the purification process was also studied so as to determine the optimum purification process. Results The optimum extraction conditions of anthocyanin from LR were as follows: the extraction solvent was 75% ethanol solution(pH3.0), extracting temperature was 40 ℃, extracting time was 2 h, and ratio of material to liquid was 1:40. Under the conditions of pH3.0, the temperature was lower than 50 ℃, and the current velocity was 1 mL/min, the maximum adsorption capacity of anthocyanin by macroporous resin reached 61.21 mg/mL, the percent recovery and content of anthocyanin were 44.35% and 118 mg/g, respectively. Black purple anthocyanin was obtained from elution solution by freeze drying. Conclusion The optimum process of extracting anthocyanin with ethanol from LR is determined, and it is proved that the AB-8 macroporous resins has a good adsorption capacity and selective adsorption. |
