目的 建立一种快速测定茶鲜叶中β-胡萝卜素的高效液相色谱(HPLC)方法。方法 以丙酮为提取溶剂; 色谱柱为SunFireTM C18 (150 mm×4.6 mm, 5 μm); 以三氯甲烷-乙腈溶液(9:1, V:V)为流动相, 等梯度洗脱; 波长450 nm; 流速1.0 mL/min; 柱温35 ℃。结果 在该色谱条件下β-胡萝卜素与其他物质得到了有效的分离, 且β-胡萝卜素在9.5×10-3~0.76 μg范围内与峰面积有良好的线性关系(r=0.99997), 回收率97.32%~101.26 %。结论 该方法重复性好、精密度高、回收率较好, 可作为茶鲜叶中β-胡萝卜素的定量测定方法。 |

Objective To develop a determination method for β-Carotene in fresh tea by high performance liquid chromatography (HPLC). Method The β-Carotene was extracted from fresh tea leaves by acetone. A SunFireTM C18 column (150 mm×4.6 mm, 5 μm) was used with the mobile phase of chloroform-acetonitrile solution (9:1, V:V). The flow rate was 1.0 mL/min, wavelength was 450 nm and the column temperature was 35 ℃. Result This method showed a good linearity (r=0.99997) for β-Carotene (ranging from 9.5×10-3 to 0.76 μg) and the recovery of β-Carotene was from 97.32% to 101.26%. Conclusion This method is precise, accurate and suitable for the determination of β-Carotene in fresh tea leaves. |
