目的 制备和筛选出一种能识别一类或几类具有结构相似的有机磷杀虫剂的广谱性鼠单克隆抗体(mAb)以对多农药残留同时进行检测。方法 制备抗有机磷组特异性单克隆抗体, 利用间接竞争性ELISA法对几种有机磷农药的标准品进行检测。结果 对乙基毒死蜱检测灵敏度较高, 最低检测限(LOD)为4 ng/mL, 对甲基对硫磷的LOD为63 ng/mL; 辛硫磷和三唑磷的检测敏感度低、一致性差。结论 根据农药化学结构的不同, 检测灵敏度有一定的差别, 对乙基毒死蜱和甲基对硫磷检测效果较好。 |

Objective To prepare and screen a broad spectrum of monoclonal antibodies (mAb), which can identify a class of organophosphorus pesticide with similar structures for the simultaneous detection of multiple pesticide residues. Methods Monoclonal antibodies against organophosphorus pesticide were prepared. Several standard of organophosphorus pesticide were detected by indirect competitive ELISA. Results The limit of detecting (LOD) value of chlorpyriphos-ethyl was 4 ng/mL with high sensitivity. The LOD value for parathion-merhyl was 63 ng/mL. The sensitivity of the detection of the phoxim and phentriazophos was low. Conclusion The sensitivity of detection was distincted because of the difference of chemical structure. The detection effect of chlorpyrifos-ethyl and parathion-methyl was good. |
