      目的 建立了一种可同时快速灵敏检测生姜中噻虫胺和噻虫嗪的量子点微球荧光免疫层析法。方法 本研究对两个项目的标记pH、抗体标记浓度及反应缓冲液体系进行了优化,通过量子点微球试纸条检测仪读取试纸条上检测线和质控线的荧光信号强度,以噻虫胺、噻虫嗪的浓度为横坐标,以检测线和质控线荧光信号强度的比值为纵坐标,建立标准曲线。结果 该方法定量检测范围噻虫胺为0.0625~8.000 μg/L、噻虫嗪为0.125~16.000 μg/L;最低检出限噻虫胺为0.032 μg/L、噻虫嗪为0.07 μg/L;加标回收率噻虫胺在84.60%~124.05%之间、噻虫嗪在88.27%~116.40%之间。与8种常见药物交叉反应率小于1%。结论 该方法具有样本前处理统一、检测简便快速、灵敏度高、可同时出具2个项目定量结果等特点,适合于大批量样品的现场筛查。

      Objective In this study, a quantum dot microsphere-based fluorescent immunochromatographic method was established for the rapid, sensitive, and simultaneous detection of clothianidin and thiamethoxam in ginger. Methods Key parameters were optimized for both analytes, including the labeling pH, antibody labeling concentration, and reaction buffer system. Fluorescence signal intensities of the test line (T-line) and control line (C-line) on the test strip were read using a quantum dot microsphere test strip detector. Standard curves were constructed with the concentrations of clothianidin and thiamethoxam as the abscissa, and the ratio of T-line to C-line fluorescence intensities as the ordinate. Results The results showed that the quantitative detection ranges were 0.0625 μg/L to 8.000 μg/L for clothianidin and 0.125 μg/L to 16.000 μg/L for thiamethoxam. The limits of detection were 0.032 μg/L for clothianidin and 0.070 μg/L for thiamethoxam. Spiked recovery rates ranged from 84.60% to 124.05% for clothianidin and 88.27% to 116.40% for thiamethoxam. Cross-reactivity rates with 8 common pesticides were less than 1%. Conclusion This method features unified sample pretreatment, simple and rapid operation, high sensitivity, and simultaneous quantitative results for two analytes, making it suitable for on-site screening of large batches of samples.
